目的:采用病、证复合模型的方法建立裸鼠阳虚证乳腺癌骨转移模型。方法:采用臀部肌肉注射氢化可的松的方法建立裸鼠阳虚证模型;采用左心室注射乳腺癌细胞MDA-MB-231悬液建立裸鼠乳腺癌骨转移模型;动态观察各组裸鼠生存情况、体重变化,进行X线摄像观察,测定环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)、血清甲状腺三碘原氨酸(T3)等值。结果:肌注氢化可的松后,裸鼠出现畏寒肢冷、活动减少、精神萎靡、皮毛少光泽、饮食减少、大便清稀等阳虚表现,且c AMP明显下降、c GMP明显升高、c AMP/c GMP值下降,T3值明显升高;左心室注射乳腺癌细胞悬液后裸鼠出现不同程度的骨转移,且经X线摄像检查确诊。结论:对裸鼠肌注氢化可的松可成功建立阳虚证模型;左心室注射法建立乳腺癌骨转移模型的成功率较高。
OBJECTIVE: To establish a bone metastasis model of breast cancer with Yang-deficiency syndrome in nude mice by means of syndrome differentiation and syndrome differentiation. Methods: The models of Yang-deficiency of nude mice were established by intramuscular injection of hydrocortisone into the buttocks. The breast cancer MDA-MB-231 suspension was injected into the left ventricle to establish the metastatic model of breast cancer in nude mice. The survival of nude mice The changes of body weight and weight were observed by X-ray. The c AMP, c GMP and T3 were determined. Results: After intramuscular injection of hydrocortisone, nude mice experienced aversion to aphrodisiac, decreased activity, apathetic, less shiny fur, decreased diet, thin stools and the like. The cAMP was significantly decreased and c GMP was significantly increased , The value of cAMP / c GMP decreased and the value of T3 increased significantly. After the left ventricular injection of breast cancer cell suspension, different degree of bone metastasis was found in nude mice and was confirmed by X-ray examination. Conclusion: The model of yang-deficiency syndrome can be successfully established by intramuscular injection of hydrocortisone in nude mice. The success rate of establishing a model of breast cancer bone metastasis by left ventricular injection is high.
