采用酶消化法培养hDPSCs,并进行鉴定。将hDPSCs在模拟微重力条件接种于PLGA支架,接种细胞数分别为0.2×106、0.4×106、0.8×106、1.2×106个/支架,以常规条件接种作为对照组。接种24h后通过细胞计数检测细胞粘附率;扫描电镜(SEM)观察细胞形态。结果:模拟微重力条件接种组随接种细胞数的增多,黏附率显著升高(P<0.01),常规条件接种组黏附率略有降低(P<0.05);当接种细胞数为0.4×106、0.8×106、1.2×106/支架时,实验组高于对照
HDPSCs were cultured by enzyme digestion and identified. HDPSCs were seeded on PLGA scaffolds under simulated microgravity conditions. The number of inoculated cells was 0.2 × 106, 0.4 × 106, 0.8 × 106 and 1.2 × 106 cells / scaffold, respectively. Cell adhesion rate was detected by cell counting 24h after inoculation; cell morphology was observed by scanning electron microscopy (SEM). Results: The rate of adhesion was significantly increased (P <0.01) and the rate of adhesion was slightly decreased in the inoculated group (P <0.05).
