目的:研究幽门螺杆菌超声粉碎抗原与霍乱毒素B亚单位组成的口服疫苗预防Hp感染的作用,探讨其保护性免疫应答的机制。方法:用Hp超声粉碎抗原2mg加CTB 10 μg作为免疫预防组,另设单纯Hp超声粉碎抗原组、单纯CTB组和PBS组为对照组。免疫4w后以活菌攻击,观察各组小鼠的免疫保护率,小鼠胃粘膜分泌性IgA抗体变化情况。结果:(1)免疫预防组小鼠免疫保护率为73.33%和对照组全部感染Hp(P <0 .0 0 1) ;(2 )免疫后4w各组小鼠未见明显的炎症反应;Hp攻击感染后4w免疫预防组小鼠可见明显的炎症反应,而各对照组相对较轻(P <0 .0 1) ;(3)Hp攻击前后免疫预防组小鼠局部胃粘膜IgA抗体显著升高(P <0 .0 1)。结论:Hp超声粉碎抗原加CTB可预防Hp感染。
Objective: To study the role of oral vaccine composed of H. pylori sonicated antigen and cholera toxin B subunit to prevent Hp infection and to explore the mechanism of its protective immune response. Methods: 2 mg Hp ultrasound pulsed antigen and 10 μg CTB were used as immunoprophylaxis group, and pure Hp ultrasound pulsed antigen group was used. CTB group and PBS group were used as control group. 4w after immunization to live bacteria to observe the immune protection rate of mice in each group mice gastric mucosa secreted IgA antibody changes. Results: (1) The immune protection rate of mice in immunoprophylaxis group was 73.33% and the control group was all infected with Hp (P <0.01). (2) No obvious inflammatory reaction was observed in mice in each group 4 h after immunization. Hp The immunosuppressive mice showed obvious inflammatory reaction at 4w after challenge, but the control group was relatively lighter (P <0.01). (3) The local IgA antibody in local immunosuppressive mice was significantly increased (P <0. 01). Conclusion: Hp ultrasound pulsed antigen plus CTB can prevent Hp infection.
