蛋白酶体降解途径是受到精密调控的特异性蛋白降解途径,在许多细胞通路中执行关键作用,但需要蛋白酶体激活因子来激活。蛋白酶体激活因子REGγ(REGγ)能显著地增强蛋白酶体的催化活性。研究发现,REGγ能以泛素非依赖方式特异地降解其靶蛋白—P21及SRC-3,突破了曾经认为REGγ/20S蛋白酶体复合物仅水解小分子多肽的认识。研究提示,REGγ具有促进细胞增殖及抗凋亡的作用,并与肿瘤发生有密切关系。REGγ/20S蛋白酶体复合物可能作为抗癌治疗的一个靶点。
The proteasomal degradation pathway is a precisely regulated, specific protein degradation pathway that performs a key role in many cellular pathways but requires proteasome activation to activate. The proteasome activator REGγ (REGγ) significantly enhances the catalytic activity of the proteasome. It was found that REGγ could specifically degrade its target protein-P21 and SRC-3 in a ubiquitin-independent manner, which broke through the once thought that the REGγ / 20S proteasome complex only hydrolyzes small molecule peptides. Research suggests that REGy can promote cell proliferation and anti-apoptosis, and is closely related to tumorigenesis. The REGy / 20S proteasome complex may serve as a target for anti-cancer therapy.
