该研究考察了性别差异对小鼠肝脏Ⅱ相代谢酶的mRNA表达的影响。将6月龄不同性别的C57BL/6小鼠分为雌、雄两组(n=6),分别取其肝脏。以荧光定量PCR(Real-time quantitative PCR,RT-qPCR)测定肝脏中II相代谢酶mRNA的相对表达量。在分析的19个II相代谢酶中,有4个代谢酶(Ugt1a1、Sult1a1、Sult2a2和Sult3a1)的mRNA水平在雌性小鼠肝脏中高表达(P<0.01或P<0.05);6个代谢酶(Ugt2b1、Ugt2b5、Ugt2b35、Sult5a1、Gstp1和Gstp2)的mRNA水平在雄性小鼠肝脏中表达较高(P<0.01或P<0.05);其他检测的II相代谢酶在雌雄小鼠肝脏中的mRNA表达没有显著性差异。性别差异显著影响小鼠肝脏中II相代谢酶的mRNA表达,这将导致药物在药效和毒性方面的个体差异。
This study examined the effects of gender differences on mRNA expression of liver phase II enzymes in mice. Six-month-old C57BL / 6 mice of different sexes were divided into female and male groups (n = 6), and their livers were harvested respectively. Real-time quantitative PCR (Real-time quantitative PCR, RT-qPCR) was used to determine the mRNA expression of phase II mRNA in the liver. The mRNA levels of four metabolic enzymes (Ugt1a1, Sult1a1, Sult2a2 and Sult3a1) were highly expressed in the liver of the female mice (P <0.01 or P <0.05) among the 19 phase II metabolizing enzymes analyzed. Six metabolic enzymes Ugt2b1, Ugt2b5, Ugt2b35, Sult5a1, Gstp1 and Gstp2) were significantly higher in the livers of male mice (P <0.01 or P <0.05). The mRNA levels of other detected phase II enzymes in the livers of both sexes No significant difference. Sex differences significantly affect the mRNA expression of phase II metabolic enzymes in the liver of mice, which leads to individual differences in drug efficacy and toxicity.
