给清醒大鼠及麻醉犬分别ⅳ异钩藤碱(Isorhy)10mg/kg和2mg/kg均使LVSP,dp/dt_(max),V_(max)等左室收缩性能指标短暂下降,而SAP,DAP及MAP呈持久性降低。在麻醉犬给药后CO,CI,HR及LVWI下降的同时SV和SI基本不变,而总外周血管阻力呈现下降趋势,反映心肌耗O_2的指标TTI明显减小。结果提示Isorhy具有短暂的抑制心肌收缩性能的作用,该作用可能参与了早期的降压机制,而Isorhy的持续降压作用主要与其扩张血管及减慢心率导致心输出量降低有关。Isorhy能减少心肌氧耗量对高血压心肌劳损可能有保护意义。
The LVSP, dp/dt_(max), V_(max) and other left ventricular contraction parameters were temporarily decreased in conscious rats and anesthetized dogs respectively iv Isorhy 10mg/kg and 2mg/kg, while SAP, DAP and MAP are persistently reduced. After administration of anesthetized dogs, CO, CI, HR and LVWI decreased while SV and SI remained unchanged, while the total peripheral vascular resistance showed a decreasing trend, indicating that the index of myocardial T2 consumption was significantly reduced. The results suggest that Isorhy has a short-term inhibitory effect on myocardial contractility, which may be involved in the early hypotensive mechanism, and Isorhy’s sustained antihypertensive effect is mainly related to its expansion of blood vessels and slowing of the heart rate leading to decreased cardiac output. Isorhy can reduce myocardial oxygen consumption may have protective significance for hypertensive myocardial injury.
