建立了用超滤 毛细管电泳技术测定血清中唾液酸含量的方法 ,并考察其最佳操作条件。该方法分离分析唾液酸可直接用紫外检测 (1 95nm) ,不需要柱前、柱后衍生。方法的回收率为 92 6%,最低检测浓度为 9 6μmol/L ,最低检测量为 39fmol。用该法测定了 30例正常人和 1 1例肺癌患者血清中唾液酸的含量 ,结果表明后者比前者有显著性增加 (P <0 0 0 1 )。用该方法测定的结果与传统使用的比色法结果之间呈良好的线性关系 (r=0 983,n =1 0 )。方法简单、灵敏 ,适合恶性肿瘤防治的基础研究和临床应用。
A method for determination of sialic acid in serum by ultrafiltration capillary electrophoresis was established and its optimum operating conditions were investigated. The method for the separation and analysis of sialic acid can be directly detected by UV (1 95nm), without column pre-column derivatization. The recovery of the method was 92.6%, the lowest detection concentration was 96μmol / L and the lowest detection was 39fmol. The method was used to determine the content of sialic acid in 30 normal subjects and 11 patients with lung cancer. The results showed that the latter had a significant increase (P <0.01). The results of this method showed a good linear relationship with the results of the conventional colorimetry (r = 0 983, n = 10). The method is simple and sensitive, suitable for the basic research and clinical application of the prevention and treatment of malignant tumors.
