目的 :观察益气活血降浊中药拮抗肾间质纤维化的作用及对结缔组织生长因子mRNA表达的影响。方法 :4 5只雄性SD大鼠随机分为假手术组、单侧输尿管结扎组及益气活血降浊中药组。分别于术后 3、6、14d摘取肾脏 ,原位杂交方法检测结缔组织生长因子mRNA的改变 ;免疫组织化学方法观察Ⅰ型胶原及α -平滑肌肌动蛋白分布及表达变化 ;Masson染色观察肾间质病变程度。结果 :与假手术组相比 ,结扎组结缔组织生长因子mRNA、Ⅰ型胶原及α -平滑肌肌动蛋白表达增强 ;肾间质病变逐渐加重 ,病变面积增加明显 ,并随天数增加而显著升高。中药组与结扎组相比 ,各项指标明显降低 ,肾间质病变面积明显缩小。结论 :在肾间质纤维化中结缔组织生长因子mRNA有着明显的上升 ,并同时伴有Ⅰ型胶原及α -SMA的升高 ,肾间质纤维化程度增加。结缔组织生长因子在肾间质纤维化形成中可能起着重要作用 ,益气活血降浊中药能够下调其mRNA的表达水平 ,对肾间质纤维化有着一定程度的拮抗作用
Objective: To observe the effect of Yiqi Huoxue Jiangzhuo on antagonizing renal interstitial fibrosis and its effect on the expression of connective tissue growth factor mRNA. Methods : Forty-five male SD rats were randomly divided into sham operation group, unilateral ureteral ligation group and Yiqi Huoxue Jiangzhuo group. The kidneys were removed on the 3rd, 6th, and 14th postoperatively respectively. In situ hybridization was used to detect the changes of connective tissue growth factor mRNA. Immunohistochemistry was used to observe the distribution and expression of type I collagen and α-smooth muscle actin. Masson staining was used to observe the kidneys. Interstitial lesions. RESULTS: Compared with the sham group, the expression of connective tissue growth factor mRNA, type I collagen and α-smooth muscle actin in the ligation group increased; the renal interstitial lesions gradually aggravated, the lesion area increased significantly, and significantly increased with the increase of the number of days. . Compared with the ligation group, the Chinese medicine group significantly reduced the indicators and significantly reduced the area of renal interstitial lesions. Conclusion : The expression of connective tissue growth factor mRNA in renal interstitial fibrosis is significantly increased, accompanied by an increase in type I collagen and α -SMA, and the degree of renal interstitial fibrosis increases. Connective tissue growth factor may play an important role in the formation of renal interstitial fibrosis. Yiqi Huoxue Jiangzhuo can down-regulate the expression level of mRNA, and it has a certain degree of antagonistic effect on renal interstitial fibrosis.
