目的建立HPLC测定消炎止痒搽剂中甲硝唑、氯霉素和水杨酸含量的方法。方法色谱柱采用YWG C18分析色谱柱(4.6 mmID×250 mm,粒径10μm);流动相为甲醇-水(50∶50,含0.3%三乙胺,磷酸调pH 3.0);流速1 ml/min;检测波长230 nm。结果甲硝唑、氯霉素和水杨酸的理论板数分别为3350、5300和6400。甲硝唑回归方程为Y=-1357265+30303746 X,r=0.9999,线性范围0.8360~3.344μg;氯霉素回归方程Y=-1137980+16799415 X,r=0.9999,线性范围1.026~4.104μg;水杨酸回归方程Y=-1209351+83566760 X,r=0.9999,线性范围0.2440~0.9760μg。平均回收率分别为99.5%(RSD 2.4%)、100.7%(RSD 1.4%)、100.3%(RSD 0.98%)。结论该法操作简便,结果准确可靠,可用于消炎止痒搽剂中甲硝唑、氯霉素和水杨酸的含量测定。
OBJECTIVE To establish a HPLC method for the determination of metronidazole, chloramphenicol and salicylic acid in anti-inflammatory and antipruritic liniment. Methods The column was YWG C18 (4.6 mmID × 250 mm, particle size 10 μm). The mobile phase was methanol-water (50:50, containing 0.3% triethylamine and phosphoric acid). The flow rate was 1 ml / min ; Detection wavelength of 230 nm. Results The theoretical plates of metronidazole, chloramphenicol and salicylic acid were 3350, 5300 and 6400 respectively. The regression equation of metronidazole was Y = -1357265 + 30303746 X, r = 0.9999, the linear range was 0.8360 ~ 3.344μg; the regression equation of chloramphenicol was Y = -1137980 + 16799415 X, r = 0.9999 with linear range of 1.026-4.104μg; Salicylic acid regression equation Y = -1209351 + 83566760 X, r = 0.9999, linear range 0.2440 ~ 0.9760μg. The average recoveries were 99.5% (RSD 2.4%), 100.7% (RSD 1.4%) and 100.3% (RSD 0.98%), respectively. Conclusion The method is simple, accurate and reliable, and can be used for the determination of metronidazole, chloramphenicol and salicylic acid in anti-inflammatory and itching liniment.
