目的为证实纯化的牛泪腺活性蛋白抗原(分子量43kD或67kD)的免疫效应及其在实验性自身免疫性泪腺炎(experimentalautoimmunedacryoadenitis,EAD)制作中的作用。方法用纯化的牛泪腺抗原及非特异性抗原植物血凝素(phytohemagglutin,PHA)作为诱导,H3-TDR掺入法检测Wistar大鼠EAD淋巴细胞转化试验。结果EAD组淋巴细胞对PHA的刺激转化反应比正常对照组偏低(P>0.05),相反对牛泪腺抗原的刺激产生高水平转化。结论牛泪腺抗原具有免疫活性,可使模型鼠泪腺致敏,动物泪腺之间存在交叉反应。
The purpose of this study was to demonstrate the immunogenicity of purified bovine lacrimal gland active protein antigen (molecular weight 43 kD or 67 kD) and its role in the development of experimental autoimmune diffuseoadenitis (EAD). Methods Purified bovine lacrimal gland antigen and non-specific antigen phytohemagglutin (PHA) were used as inducers to detect the EAD lymphocyte transformation in Wistar rats by H3-TDR incorporation method. Results The response of PHA stimulated by lymphocytes in EAD group was lower than that in normal control group (P> 0.05). On the contrary, stimulation of cattle lacrimal gland antigen produced a high level of transformation. Conclusions Bovine lacrimal gland antigen is immunologically active, which can sensitize the lachrymal gland of mice and cause cross-reaction between lacrimal gland of animals.
