目的:探究黄精多糖(Polygonatum sibiricum polysaccharides,PSP)对H9c2心肌细胞缺氧/复氧(hypoxia-reoxygenation,H/R)损伤的保护作用。方法:体外无菌培养H9c2心肌细胞,制备缺氧/复氧损伤模型。应用A0-EB和Hoechst33324染色法观察心肌细胞凋亡形态学变化,流式细胞仪检测细胞凋亡率,采用MTT法检测心肌细胞存活率,Westen Blot法检测H9c2心肌细胞Caspase-3、Bcl-2及Bax蛋白表达。结果:缺氧/复氧模型组中H9c2细胞存活率明显降低,细胞核浓染致密、固缩,细胞凋亡率明显增加,Caspase-3的表达和Bax/Bcl-2比率显著上调;与模型组比较,黄精多糖1.0、1.5、2.0mg/ml药物组中细胞存活率均明显升高,细胞核淡染,轻微缩小,细胞凋亡率降低,Bax/Bcl-2比率明显下调,Caspase-3表达水平被显著抑制。结论:黄精多糖对H/R诱导的H9c2心肌细胞具有保护作用,其作用可能与抑制心肌细胞凋亡有关。
Objective: To investigate the protective effect of Polygonatum sibiricum polysaccharides (PSP) on hypoxia-reoxygenation (H / R) injury in H9c2 cardiomyocytes. Methods: H9c2 cardiomyocytes were aseptically cultured in vitro and hypoxia / reoxygenation injury models were established. The morphological changes of cardiomyocytes were observed by A0-EB and Hoechst33324 staining. The apoptosis rate of cardiomyocytes was detected by flow cytometry. The survival rate of cardiomyocytes was detected by MTT assay. Caspase-3, Bcl-2 And Bax protein expression. Results: The survival rate of H9c2 cells in hypoxia / reoxygenation model group was significantly lower than that in model group (P <0.05) Compared with the control group, the cell viability in the 1.0, 1.5 and 2.0 mg / ml Polygonatum polysaccharides groups were significantly increased, the cell nucleus was lightly stained and slightly reduced, the cell apoptosis rate was decreased, the ratio of Bax / Bcl-2 was significantly decreased and the expression of Caspase-3 Was significantly suppressed. CONCLUSION: Polygonatum polysaccharide can protect H9c2 cardiomyocytes induced by H / R, which may be related to the inhibition of cardiomyocyte apoptosis.
