目的 探讨抗骨增生胶囊对去卵巢大鼠骨质疏松的防治作用.方法 雌性SD大鼠40只,随机分为假手术组、模型组、雌激素组和抗骨增生胶囊组.行双侧卵巢切除术复制骨质疏松模型.手术后1周,假手术组与模型组均生理盐水灌胃,雌激素组皮下注射17-β雌二醇,抗骨增生组给予抗骨增生胶囊灌胃,给药持续时间12周.观察抗骨增生胶囊对去卵巢所致大鼠骨密度、骨组织形态学的影响,采用ELISA法检测血清骨钙素、Ⅰ型前胶原N端前肽水平,Western-blot检测股骨骨钙素(OPG)和核因子NF-kB受体激活因子配体(RANKL)蛋白表达.结果 模型组大鼠骨密度低于假手术组,骨小梁减少且排列稀疏,骨小梁间距增宽,血清骨钙素(BGP)、Ⅰ型前胶原N端前肽(PINP)水平上升,骨皮质OPG蛋白表达下降,RANKL蛋白表达升高;雌激素组和抗骨增生胶囊组均能改善骨组织形态学改变,降低血清BGP、PINP水平,提高OPG蛋白和降低RANKL蛋白表达.结论 抗骨增生胶囊可能通过降低骨代谢高转换水平,提高OPG蛋白和降低RANKL蛋白表达,达到防治骨质疏松目的.“,”Objective To investigate the preventive and therapeutic effects of Kangguzengsheng Capsues on osteoporosis in ovariectomized rats.Methods The 40 female SD rats were randomly divided into 4 groups:Sham group,OVX group,OVX + E group and OVX + KG group.The osteoporotic models were established by bilateral ovariectomy.One week after ovariectomy,the sham and OVX group were given with physiological saline solution by intragastric gavage;in the OVX + E group,17β-E2 was injected subcutaneously;Kangguzengsheng Capsules solution was given by intragastric administration in the OVX + KG group.After treatment for 12w,the changes of bone mineral density and cortical morphology were observed;the levels of BGP and PINP in plama were measured by ELISA;and the expression of OPG and RANKL in bone tissue was detected with Western blot.Results Compared with the sham group,BMD of OVX group was decreased significantly,bone trabecular arranged sparsely and irregularly.The trabecular became thinner,trabecular separation became wider;serum BGP,PINP levels were significantly increased;the protein expression of OPG significantly decreased,and RANKL protein expression was significantly increased.For the OVX + E and OVX + KG group,the morphological changes and BMD were improved significantly than that of OVX group;the serum level of BGP,PINP decreased;the protein expression of OPG was increased and that of RANKL was decreased.Conclusion Kangguzengsheng Capsules can prevent osteoporosis in ovariectmized rats by reducing bone turnover,increasing OPG and reducing RANKL expression.
