用分光光度酶偶联速率法,测定用过氧钒烟酸络合物(POV)和二甲双胍(MET)治疗前后糖尿病鼠肝细胞质的磷酸烯醇型丙酮酸羧化酶(PEPCK)活性。结果显示:糖尿病鼠肝细胞质的PEPCK活性较正常鼠明显增加(P<0.01);POV治疗组经POV治疗后该酶活性明显受抑制(P<0.01),而MET治疗组经MET治疗后该酶活性虽有轻微降低但无统计学意义。POV和MET对正常大鼠的PEPCK均无明显影响。此结果为POV降低高血糖的分子生化机理提供理论依据,亦为探讨治疗糖尿病新的降糖药提供实验基础。
The activity of phosphoenolpyruvate carboxylase (PEPCK) in the liver cytoplasm of diabetic mice before and after treatment with peroxovanadate complexes (POV) and metformin (MET) was measured by spectrophotometric enzyme coupling rate assay. The results showed that the activity of PEPCK in hepatic cytoplasm of diabetic rats was significantly increased compared with that of normal rats (P <0.01). The activity of PEPCK in POV treated group was significantly inhibited by POV treatment (P <0.01) Although after treatment, the enzyme activity decreased slightly but not statistically significant. POV and MET had no significant effect on PEPCK in normal rats. The results provide a theoretical basis for POV to reduce the molecular mechanism of hyperglycemia and provide experimental basis for exploring new hypoglycemic agents for treating diabetes.
