目的探讨三维适形调强放疗(intensity modulated radiation therapy,IMRT)的放射线对鼠骨细粒棘球蚴生发细胞生长及凋亡的影响。方法骨细粒棘球蚴病子午沙鼠20只,将骨细粒棘球蚴内囊中的生发细胞进行分离,并进行体外培养,对体外培养成功的骨细粒棘球蚴生发细胞分为IMRT放疗组和空白对照组。IMRT放疗组给予总剂量为40Gy的IMRT放疗,对照组仅给予假照射。于放疗结束24h后采用流式细胞仪检测骨细粒棘球蚴生发细胞凋亡率,采用Western blot法检测骨棘球蚴生发细胞中凋亡相关蛋白caspase-3蛋白表达情况,采用RT-PCR法定量检测骨棘球蚴生发细胞caspase-3mRNA表达水平。结果 IMRT作用24h后,IMRT放疗组骨细粒棘球蚴生发细胞凋亡率、caspase-3蛋白及mRNA表达灰度值((67.21±14.40)%、7.761±1.447、59.44±19.28)明显高于对照组((2.91±0.51)%、0.129±0.054、1.17±0.50),差异均有统计学意义(P<0.05)。结论 IMRT对骨细粒棘球蚴生发细胞生长有明显诱导凋亡作用,其作用机制可能是通过调节凋亡相关蛋白caspase-3而实现的。
Objective To investigate the effects of three-dimensional intensity modulated radiation therapy (IMRT) on the growth and apoptosis of germinal cells of Echinococcus granulosus. Methods Twenty gerbilular gerbils of Echinococcus granulosus were isolated and the germinal cells in the inner capsule of Echinococcus granulosus were isolated and cultured in vitro. The germinal cells of Echinococcus granulosus IMRT radiotherapy group and blank control group. IMRT radiotherapy group was given a total dose of 40Gy IMRT radiotherapy, the control group was given only fake irradiation. The apoptotic rate of germinal cells of Echinococcus granulosus was detected by flow cytometry 24h after radiotherapy, and the expression of caspase-3 protein in germ cell of Echinococcus granulosus was detected by Western blot. Quantitative detection of sporozoite gerbil cells caspase-3mRNA expression levels. Results After treated with IMRT for 24 h, the apoptotic rate, the gray value of caspase-3 protein and mRNA expression (67.21 ± 14.40%), 7.761 ± 1.447 and 59.44 ± 19.28 in IMRT group were significantly higher than those in IMRT group (2.91 ± 0.51)%, 0.129 ± 0.054, 1.17 ± 0.50 respectively), the differences were statistically significant (P <0.05). CONCLUSION: IMRT can obviously induce the apoptosis of germinal cells of Echinococcus granulosus. The possible mechanism is that IMRT regulates apoptosis-related protein caspase-3.
