目的研究了仿刺参糖氨聚糖(HGAG)对小鼠特异性免疫功能的影响,探讨其对小鼠的特异性免疫的作用。方法不同浓度的HGAG与小鼠脾细胞悬液共培养44h,MTT法检测;将昆明种小鼠32只,分为4组,每组8只,空白组注射生理盐水,HGAG组(0.1,1,10mg·kg-1),每天给药,第1天将全部小鼠腹腔注射压积SRBC,4d后观察小鼠足趾肿胀程度;5d后小鼠摘眼球取血,540nm测OD值,计算半数溶血值(HC50);将小鼠处死,取脾制成单细胞悬液经培养,413nm测OD值,检测抗体生成细胞数。结果 HGAG在0.05~25μg·mL-1范围内能明显促进小鼠脾淋巴细胞的增殖,与空白组相比具有显著性差异(P<0.05),在0.5~10μg·mL-1范围内,活性优于阳性组(P<0.05),在HGAG浓度为1μg·mL-1时促进增殖作用最为明显;HGAG在低、中、高剂量组均能促进小鼠迟发性变态反应,且随着剂量浓度的增加而增高(P<0.05);显著提高血清溶血素及抗体生成细胞数(P<0.05)。HGAG对正常小鼠的细胞免疫、体液免疫均有明显的增强作用,进一步说明HGAG在特异性免疫方面具有一定的调节作用。
Objective To study the effect of HGAG on the specific immune function of mice and to explore its effect on the specific immunity in mice. Methods HGAG with different concentrations were co-cultured with mouse spleen cell suspension for 44h and detected by MTT method. Thirty-two Kunming mice were divided into 4 groups with 8 rats in each group. The rats in the blank group were injected with normal saline, HGAG group (0.1, , 10mg · kg-1) every day. All mice were intraperitoneally injected with SRBC on day 1, and the degree of toe swelling in mice was observed 4 days later. Half the value of hemolysis (HC50); the mice were sacrificed, spleen made of single cell suspension culture, 413nm measured OD value, the number of antibody-producing cells. Results HGAG could significantly promote the proliferation of splenic lymphocytes in the range of 0.05 ~ 25μg · mL-1, which was significantly different from the blank group (P <0.05). The activity of HGAG in the range of 0.5-10μg · mL-1 (P <0.05), and the most obvious effect was promoted when the concentration of HGAG was 1μg · mL-1. HGAG could promote delayed-type hypersensitivity in low, medium and high dose groups, and with the dose (P <0.05), and significantly increased the number of hemolysin and antibody-producing cells (P <0.05). HGAG on normal mice, cellular immunity, humoral immunity were significantly enhanced, further demonstrating that HGAG in the specific immune regulation has a certain role.
