为研究金雀异黄素对人胃腺癌细胞SGC 790 1增殖、DNA合成、一氧化氮产生量、一氧化氮合酶活性及其对诱导型一氧化氮合酶 (inducednitricoxidesynthase ,iNOS)基因表达的影响 ,应用MTT、3 H TdR掺入、DNA琼脂糖电泳、分光光度法、免疫组化及RT PCR方法 ,研究Gen抑制癌细胞生长的机制。结果表明Gen抑制SGC 790 1细胞的增殖、DNA合成 ,诱导细胞凋亡 ,并可显著诱导iNOSmRNA转录、增加iNOS蛋白表达 ,促进一氧化氮产生 ,提高一氧化氮合酶活性。为将Gen应用于胃腺癌的化学防治提供了实验依据
To investigate the effects of genistein on the proliferation, DNA synthesis, nitric oxide production, nitric oxide synthase activity and the expression of inducible nitric oxide synthase (iNOS) gene in human gastric adenocarcinoma cell SGC7901 The effects of Gen on the growth of cancer cells were investigated by MTT, 3 H TdR incorporation, DNA agarose electrophoresis, spectrophotometry, immunohistochemistry and RT-PCR. The results showed that Gen inhibited SGC7901 cell proliferation, DNA synthesis, induced apoptosis, and could significantly induce iNOS mRNA transcription, increase iNOS protein expression, promote nitric oxide production and increase nitric oxide synthase activity. It provided the experimental basis for the chemical control of Gen in gastric adenocarcinoma
