目的观察薏苡仁酯对结肠癌SW480增殖和凋亡的影响。方法结肠癌SW480细胞分为4组,其中A组为对照组,B,C,D组分别在培养基中给予5,10,20mg/L薏苡仁酯处理。采用噻唑蓝比色法检测4组细胞生长情况,流式细胞术检测细胞周期和凋亡情况。结果作用24h后,B,C,D组的抑制率分别为16.43%,22.51%,31.24%,呈剂量依赖关系,与A组比较差异均有统计学意义(P<0.05);给药后24hD组细胞G0~G1期比例较A组增高,S期细胞比例较A组降低(P<0.05);D组24,48h凋亡率明显高于A组,差异有统计学意义(P<0.05)。结论薏苡仁酯可在体外直接抑制结肠癌细胞的增殖,诱导凋亡,在结肠癌中药防治中有一定作用。
Objective To observe the effect of Coixenolide on the proliferation and apoptosis of colon cancer SW480 cells. Methods Colon cancer SW480 cells were divided into 4 groups, among which, group A was the control group, group B, C and D were treated with 5, 10 and 20 mg / L Coixenolide respectively. The growth of 4 groups of cells was detected by thiazolyl blue colorimetric assay. The cell cycle and apoptosis were detected by flow cytometry. Results After 24h, the inhibitory rates of group B, C and D were 16.43%, 22.51% and 31.24%, respectively, in a dose-dependent manner. The difference was statistically significant compared with group A (P <0.05) The percentage of G0-G1 phase cells in group D was higher than that in group A, and the percentage of cells in S phase was lower than that in group A (P <0.05). The apoptosis rates in group D at 24 and 48 h were significantly higher than those in group A (P <0.05) . Conclusions Coixenolide can directly inhibit the proliferation and induce apoptosis of colon cancer cells in vitro, which may play a role in the prevention and treatment of colon cancer.
