目的:研究癌基因HER-2、端粒酶(the human telomerase gene,hTERC)在不同程度子宫颈病变脱落细胞中的表达,以探讨其在子宫颈癌发生、发展中的作用及临床意义。方法:采用荧光原位杂交(fluorescent in situ-hybridization,FISH)方法,检测15例正常、76例子宫颈上皮内瘤变(CIN)及24例浸润性子宫颈癌脱落细胞中HER-2、hTERC的表达情况。结果:HER-2在正常子宫颈脱落细胞、CINⅠ、CINⅡ~Ⅲ及浸润性子宫颈癌中的阳性表达率分别为0.0%、6.3%、16.6%及58.3%,hTERC基因阳性表达率为6.7%、18.8%、65.0%及100.0%。②HER-2、hTERC基因在CINⅠ、CINⅡ~Ⅲ中阳性表达率比较,差异无显著性(P>0.05);在CINⅡ~Ⅲ及子宫颈癌组阳性表达率均显著高于正常组(P<0.05),同时子宫颈癌组明显高于CIN组(P<0.05)。③HER-2高表达与细胞分级有明显相关性(P<0.05),与临床分期、病理类型无关。结论:HER-2、hTERC基因高表达在子宫颈癌发生、发展中可能起重要作用,在子宫颈病变脱落细胞中检测HER-2、hTERC基因可以作为子宫颈癌早期筛查、病情监测和评估预后的辅助指标。
Objective: To investigate the expression of oncogene HER-2 and hTERC in cervical exfoliated cells with varying degrees of cervical cancer to investigate its role in the occurrence and development of cervical cancer and its clinical significance.
