目的研究低剂量铅对原代培养的海马神经元神经细胞黏附分子(NCAM)表达的影响。方法将孕18dWistar大鼠麻醉后剖腹取出胎鼠,建立原代培养的海马神经元,与不同剂量的氯化铅(PbCl2)共孵育后,应用免疫印迹法技术检测培养第1至7天海马神经元NCAM的表达。结果正常条件下培养的海马神经元,第1天NCAM的表达极其微弱,积分吸光度值为14,NCAM表达的高峰时间在培养第3~5天,积分吸光度值为2542~2580;随培养时间的延长,NCAM表达逐渐降低。在铅的作用下,NCAM的表达在培养早期(第2~4天)受到抑制,且呈现明显的剂量效应关系,随时间延长,抑制程度逐渐减弱;培养至第5天,铅的抑制作用消失,10-2、10-3及10-4mmol/LPbCl2剂量组NCAM的表达甚至超出了对照组;此后,所有组的NCAM表达出现下降趋势,铅对NCAM表达的抑制又表现出剂量效应关系。结论低剂量铅暴露显著抑制了原代培养的大鼠海马神经元NCAM的表达,并造成了海马神经元NCAM表达时程上的延迟。
Objective To study the effect of low dose lead on the expression of neural cell adhesion molecule (NCAM) in primary cultured hippocampal neurons. Methods Pregnant 18-day Wistar rats were anesthetized and fetuses were removed by cesarean section. Primary cultured hippocampal neurons were established and co-incubated with different concentrations of lead chloride (PbCl2). Western blotting was used to detect the hippocampal neurons Metamorphic NCAM expression. Results The NCAM expression was extremely weak on day 1, and the integral absorbance value was 14. The peak time of NCAM expression was between day 3 and day 5, and the integral absorbance value was 2542 ~ 2580. With the increase of culture time Prolonged, NCAM expression gradually decreased. Under the action of lead, the expression of NCAM was inhibited in the early stage of culture (days 2 to 4), and showed a dose-dependent effect. Conclusion Low-dose lead exposure significantly inhibits NCAM expression in primary cultured hippocampal neurons and delays the time course of NCAM expression in hippocampal neurons.
