目的探讨青蒿琥酯增敏对宫颈癌HeLa细胞裸鼠移植瘤凋亡的影响。方法建立裸鼠移植瘤模型,待移植瘤体积平均增至大约(5 mm×5 mm×5 mm),将裸鼠随机分为对照组、单药组、单照组和联合组,单药组和联合组每只裸鼠给予青蒿琥酯100 mg.kg~(-1).d~(-1),对照组和单照组每只裸鼠给予等体积生理盐水,处理7 d,第7 d给药后单照组和联合组立即给予一次性~(60)Coγ射线10Gy照射,照后观察14 d,14 d后剥瘤,TUNEL法检测移植瘤组织中细胞的凋亡。结果联合组裸鼠移植瘤瘤质量较单照组明显减小[(0.64±0.11)g vs(1.31±0.58)g](P<0.05),抑瘤率达71.17%。联合组移植瘤组织中细胞凋亡数较单照组显著增大[(77.5±8.07)vs(48.80±6.71)](P<0.05)。结论青蒿琥酯能明显增加HeLa细胞裸鼠移植瘤对射线的放射敏感性,作用机制与其增强射线诱导移植瘤组织中细胞凋亡有关。
Objective To investigate the effect of artesunate on apoptosis of transplanted cervical cancer HeLa cells in nude mice. Methods The nude mice model of transplantation was established. When the volume of the transplanted tumor increased to about 5 mm × 5 mm × 5 mm, the nude mice were randomly divided into control group, monotherapy group, monotherapy group and combination group. In the control group and the single group, the same volume of normal saline was given to each nude mouse in the combination group for 7 days. The nude mice in the combination group received artesunate 100 mg.kg -1 .d -1, After administration for 7 days, the single irradiation group and the combination group were given 10Gy irradiation of 60Coγ ray immediately. The cells were observed for 14 days and 14 days after the irradiation. TUNEL assay was used to detect the apoptosis in the transplanted tumor tissue. Results Compared with the control group, the tumor mass of nude mice in the combination group was significantly decreased ([0.64 ± 0.11] g vs (1.31 ± 0.58) g] (P <0.05), and the inhibition rate was 71.17%. The number of apoptotic cells in the transplanted tumor tissue in combination group was significantly higher than that in the control group [(77.5 ± 8.07) vs (48.80 ± 6.71)] (P <0.05). Conclusion Artesunate can significantly increase the radiosensitivity of HeLa cells to xenografts in nude mice and its mechanism is related to the enhancement of apoptosis induced by radiation in xenograft tumors.
