目的:探讨蓝光诱导对蛹虫草Cordyceps militaris L.菌丝体和子实体中胞内多糖的单糖组成以及虫草素和虫草多糖含量的影响。方法:以蛹虫草为材料,使用气相色谱检测蓝光照射144 h后菌丝体及子实体中多糖的单糖组成;并在蓝光照射下的不同时间段分别取样,以检测菌丝体及子实体中虫草素和虫草多糖的含量。同时观察蛹虫草菌丝体和子实体在蓝光照射诱导过程中的生长情况。结果:蛹虫草菌丝体多糖是一种含有4种单糖的杂多糖;子实体多糖是一种含有5种单糖的杂多糖。在菌丝体多糖的检测试验中,无论是蓝光照射还是黑暗培养,两组的多糖含量变化规律几乎相似,都是在一定的时间段开始发生变化。在子实体多糖的检测中,无论是蓝光照射还是白光对照,两组多糖含量的变化规律也基本相同。在菌丝体以及子实体虫草素的检测中,虫草素含量在两组实验中均呈现出明显的上升趋势。结论:蓝光诱导对蛹虫草胞内多糖中单糖的组成种类和数量有一定的影响。而且蓝光诱导对蛹虫草中虫草素和虫草多糖含量的提高有一定的促进作用。蓝光有利于蛹虫草的形态建成,促进子实体的分化和生长发育。该项研究首次报道了蓝光诱导对蛹虫草多糖中单糖组成的种类和数量的影响,为进一步研究蓝光诱导对蛹虫草活性物质的代谢奠定基础。
OBJECTIVE: To investigate the effect of blue light on the monosaccharide compositions of intracellular polysaccharides and the contents of polysaccharides of Cordyceps and Cordyceps militaris in Cordyceps militaris L. mycelia. Methods: Using Cordyceps militaris as material, the monosaccharide composition of polysaccharides in mycelium and fruiting body was detected by gas chromatography after 144 h of blue light irradiation. The mycelium and fruiting bodies were sampled at different time periods under blue light irradiation Cordycepin and Cordyceps polysaccharide content. The growth of mycelium and fruiting bodies of Cordyceps militaris were observed during the induction of blue light. Results: Cordyceps militaris mycelium polysaccharide is a heteropolysaccharide containing four kinds of monosaccharides; fruiting polysaccharides is a heteropolysaccharide containing five kinds of monosaccharides. In the mycelium polysaccharide test, either the blue light or dark culture, the two groups of polysaccharide content changes are almost similar, are beginning to change in a certain period of time. In the detection of fruiting body polysaccharides, whether the blue light or white light control, two groups of polysaccharide content changes are basically the same. In mycelia and fruiting body cordycepin detection, cordycepin content showed a clear upward trend in both groups. Conclusion: The induction of blue light on Cordyceps militaris intracellular polysaccharides monosaccharide composition types and quantity have a certain impact. Moreover, blue light induced Cordyceps Militaris and Cordyceps polysaccharide content increased to some extent. Blu-ray is conducive to Cordyceps militaris morphogenesis, promote fruiting body differentiation and growth and development. This study reports for the first time the effects of blue light on the type and amount of monosaccharide in the polysaccharides of Cordyceps militaris, and lays the foundation for further study on the blue light-induced metabolism of active substances in Cordyceps militaris.
