目的通过观察家兔系膜增殖型肾炎(MSPGN)血内皮素(ET-1)和降钙素基因相关肽(CGRP)水平的变化,探讨肾舒胶囊治疗MSPGN的作用机制。方法应用生物化学、放射免疫和自动图像分析的方法。检测24小时尿蛋白的排泄量,血浆ET-1及CGRP及肾组织系膜基质面积的变化。结果MSPGN家兔24小时尿蛋白排泄量升高,ET-1升高,CGRP降低,系膜基质面积增大与肾舒胶囊治疗组相比具有显著性差异(P<0.05)。结论MSPGN家兔ET-1升高CGRP降低,系膜基质面积增宽。肾舒胶囊通过升高CGRP降低ET-1,降低肾小球系膜基质面积对MSPGN有明显保护作用。
Objective To investigate the mechanism of Shenshu Capsule in the treatment of MSPGN by observing the changes of blood endothelin (ET-1) and calcitonin gene-related peptide (CGRP) levels in rabbits with mesangial proliferative nephritis (MSPGN). Methods Biochemistry, radioimmunoassay, and automated image analysis methods were used. The 24-hour urinary protein excretion, plasma ET-1 and CGRP, and renal tissue mesangial matrix area were measured. Results The 24-hour urinary protein excretion of MSPGN rabbits increased, ET-1 increased, CGRP decreased, and the area of mesangial matrix increased compared with Shenshu capsule treatment group (P<0.05). Conclusion MSPGN rabbit ET-1 increased CGRP level and increased the area of mesangial matrix. Shen Shu Capsule reduced ET-1 by increasing CGRP, and decreased glomerular mesangial matrix area had obvious protective effect on MSPGN.
