尿激酶(urokinase,UK)是由肾脏产生,存在于尿中的纤溶酶原激活剂,因无种属特异性、无抗原性和毒性小而成为临床溶血栓治疗的药物之一,从新鲜人尿中提取的尿激酶,主要有 54 000(HUK)和 33 000(LUK)两种,目前衡量尿激酶纯度仍以HUK含量为指标,主要用电泳方法检测。应用卫生部药品标准提供的方法进行检测,样品的分离不理想。我们应用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE),比较了不同的样品处理方法和不同的电泳系统对UK纯度检测的影响,建立了一种简便实用的UK纯度检测方法。
Urokinase (urokinase, UK) is produced by the kidneys and exists in the urine of plasminogen activator, because no species-specific, non-antigenic and toxic to become one of the clinical thrombolytic drugs, from fresh Urokinase extracted from human urine, mainly 54 000 (HUK) and 33 000 (LUK) two, the current measurement of uricase purity is still HUK content as an indicator, mainly by electrophoresis. Application of the method provided by the Ministry of Health drug standards for testing, sample separation is not satisfactory. We used SDS-polyacrylamide gel electrophoresis (SDS-PAGE) to compare the effects of different sample processing methods and different electrophoresis systems on the UK purity test, and established a simple and practical UK purity test method.
