目的研究成纤维细胞与结肠癌细胞相互作用对二者表达细胞外基质金属蛋白酶诱导因子(EMMPRIN)的影响,初步探讨肿瘤-基质相互作用在结肠癌侵袭转移中的作用。方法结肠癌 SW480细胞与 HELF 成纤维细胞以 RPMI 1640培养液分别共培养0、12、24、48 h,通过 RT-PCR 和免疫细胞化学方法检测 SW480和 HELF 细胞中 EMMPRIN 的表达。结果共培养组 SW480细胞EMMPRIN mRNA 和蛋白的表达均明显升高,HELF 细胞本来不表达 EMMPRIN,与 SW480细胞共培养12、24、48 h 后检测到 EMMPRIN 表达,且随时间延长而表达增加。结论成纤维细胞与结肠癌细胞相互作用上调 SW480细胞 EMMPRIN 的表达,并诱导 HELF 细胞表达 EMMPRIN,有可能在结肠癌侵袭转移中发挥重要作用。
Objective To investigate the effect of the interaction between fibroblasts and colon cancer cells on the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) and the role of tumor-matrix interactions in the invasion and metastasis of colon cancer. Methods Colorectal cancer SW480 cells and HELF fibroblasts were co-cultured with RPMI 1640 medium for 0, 12, 24 and 48 h respectively. The expression of EMMPRIN in SW480 and HELF cells was detected by RT-PCR and immunocytochemistry. Results The expression of EMMPRIN mRNA and protein in SW480 cells was significantly increased in co-cultured group, but not in HELF cells. EMMPRIN was detected in 12, 24, and 48 hours after co-cultured with SW480 cells, and the expression of EMMPRIN was detected with prolonged time. Conclusion The interaction between fibroblasts and colon cancer cells may up-regulate the expression of EMMPRIN in SW480 cells and induce the expression of EMMPRIN in HELF cells, which may play an important role in the invasion and metastasis of colon cancer.
