观察缺氧缺血性脑损伤 (HIBD)新生大鼠长时间吸高浓度氧后脑病理学及DNA损伤的变化。 HIBD新生大鼠吸纯氧 48小时 ,观察脑大体和光镜下的变化 ,并进行脑组织 DNA电泳。结果 HIBD大鼠吸氧后 5天与未吸氧组比 ,左脑丢失重量减少、左脑大体病变程度减轻 ,但均无统计学意义 (P均 >0 .0 5 ) ;吸氧组死亡率高于未吸氧组 ,分别为 11.5 %和 4.2 % ,差异亦无显著性 (P均 >0 .0 5 ) ;HIBD大鼠吸氧后 5天 ,光镜下左脑损伤程度较未吸氧组稍轻 ,但未能进行统计学处理 ;DNA电泳结果表明 ,缺氧缺血后 48小时皮质、海马、纹状体和丘脑的 DNA电泳在模糊涂片带的基础上均可见“梯形条带”,表明既有细胞坏死 ,又有细胞凋亡存在。缺氧缺血后再吸氧 48小时相同脑区的 DNA电泳与未吸氧组相比无明显差别。结论 HIBD新生大鼠吸纯氧 48小时后脑病理及 DNA损伤无明显改善 ,死亡率反而有所增加
To observe the changes of cerebral pathology and DNA damage in neonatal rats with hypoxic-ischemic brain damage (HIBD) after long-term high concentration oxygen inhalation. HIBD neonatal rats were inhaled oxygen for 48 hours, observed changes in the brain and light microscope, and brain DNA electrophoresis. Results Compared with the non-oxygen group, the weight loss of left brain and the severity of left brain lesion in HIBD rats at 5 days after oxygen inhalation were not statistically significant (all P> 0.05) Which were 11.5% and 4.2% respectively higher than that of non-oxygen inhalation group (P> 0.05). In the fifth day after oxygen inhalation, the degree of left brain injury in the HIBD rats was lower than that of the control group The results of DNA electrophoresis showed that DNA electrophoresis of cortex, hippocampus, striatum and thalamus 48 hours after hypoxia-ischemia could be seen on the basis of fuzzy smear zone. “Trapezoidal band ”, Indicating that both the cell necrosis, there are apoptosis. Hypoxic-ischemic reoxygenation 48 hours after the same brain DNA electrophoresis compared with no oxygen group no significant difference. Conclusion HIBD neonatal rats with pure oxygen for 48 hours had no significant improvement in brain pathology and DNA damage, but the mortality rate increased
