目的为提高食用油中黄曲霉毒素ELISA检测的精密度与准确度,对检测相关条件进行优化探讨。方法通过调整样品前处理以及免疫反应过程中一些实验参数,对比不同实验条件下实际样品测定值的RSD与回收率,取一定量样品提取液加入样品稀释液后按ELISA测定程序进行测定。结果在优化条件下样品加标回收率在75.5%~115.6%之间,同一个样品提取液做多孔平行其RSD小于8.5%。以超高效液相色谱串联质谱(UPLC-MS-MS)进行确认检测结果满意。结论优化后的检测条件有效地改善了检测过程中不确定性,样品检测结果的平行性和重复性较好,本实验对食用油中黄曲霉毒素B1的有关优化研究参数可以作为实际检测分析的参考,适用于实验室常规检测。
Objective To improve the precision and accuracy of aflatoxin ELISA in food oil, and to optimize the detection conditions. Methods By adjusting the sample pretreatment and some experimental parameters in the course of immune reaction, the RSD and recovery of the actual samples under different experimental conditions were compared. A certain amount of sample extract was added to the sample diluent and then measured by ELISA. Results The spiked recoveries of spiked samples ranged from 75.5% to 115.6% under the optimized conditions. The RSD of the same sample was less than 8.5%. Confirmation by ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS-MS) showed satisfactory results. Conclusion The optimized detection conditions can effectively improve the uncertainty of the detection process. The parallelism and repeatability of the detection results are good. The parameters of optimization of aflatoxin B1 in edible oils can be used as the actual detection and analysis Reference, suitable for routine laboratory testing.
