目的:探究鬼针草提取物在急性胰腺炎大鼠肺损伤中的保护作用。方法:随机将大鼠分为8组:对照(Sham)组,模型(SAPLI)组,鬼针草提取物低(50mg/kg)、中(100mg/kg)、高(200mg/kg)剂量组,鬼针草提取物+TGF-β1组(外源性TGF-β1),鬼针草提取物+empty vector组、鬼针草提取物+Angptl4plasmid vector组。大鼠实验结束后12h,检测各组肺湿干重比(W/D),血清淀粉酶(AMY)活性,肺组织髓过氧化物酶的活性(MPO),炎性因子TNF-α、IL-6、IL-1β的分泌水平,RT-PCR和western blot分别检测TGF-β1、Smad3、Angptl4的mRNA和蛋白表达水平。结果:和Sham组比较,SAPLI组AMY、MPO活性、肺湿干重比、TNF-α、IL-6、IL-1β的水平和TGF-β1、Smad3、Angptl4的表达水平显著升高,而鬼针草提取物可使上述指标下降(P<0.05);注射外源性TGF-β1蛋白或过表达Angptl4可逆转TFB的肺损伤保护作用。结论:鬼针草提取物对急性胰腺炎大鼠相关性肺损伤有保护作用,这与阻断TGF-β1/Smad3/Angptl4信号通路有关。
Objective: To investigate the protective effect of Bidens extract on lung injury in rats with acute pancreatitis. Methods: The rats were randomly divided into 8 groups: sham group, model group (SAPLI), Bidens officinalis extract low (50mg / kg), medium (100mg / kg) and high (200mg / kg) , Bidens extract + TGF-β1 group (exogenous TGF-β1), Bidens extract + empty vector group, Bidens extract + Angptl4plasmid vector group. The rat lung wet weight ratio (W / D), serum amylase (AMY) activity, lung tissue myeloperoxidase activity (MPO), inflammatory cytokines TNF-α, IL -6 and IL-1β, respectively. The mRNA and protein levels of TGF-β1, Smad3 and Angptl4 were detected by RT-PCR and western blot respectively. Results: Compared with Sham group, the levels of AMY and MPO, the wet / dry weight ratio of lung, the levels of TNF-α, IL-6 and IL-1β and the expressions of TGF-β1, Smad3 and Angptl4 in SAPLI group were significantly increased The above indexes decreased (P <0.05). The injection of exogenous TGF-β1 protein or overexpression of Angptl4 could reverse the protective effect of TFB on lung injury. Conclusion: Bidens extract has a protective effect on lung injury in rats with acute pancreatitis, which is related to blocking TGF-β1 / Smad3 / Angptl4 signaling pathway.
