目的观察AML1B基因及其异构体AML1A对靶基因启动子转录活性的影响,探讨造血干细胞定向分化和白血病发生的机制。方法构建AML1A和AML1B的表达质粒,与含有靶基因巨噬细胞集落刺激因子受体(M-CSF-R)启动子的荧光素酶报告质粒共转染非洲绿猴肾CV-1细胞,测定荧光素酶活性,分析AML1A及AML1B对靶基因转录活性的影响。结果AML1B对M-CSF-R具有明显的转录激活作用,这种作用具有明显的序列特异性和剂量依赖性;AML1A无转录激活作用,而且拮抗AML1B,使M-CSF-R的表达水平明显下降。结论AML1转录本结构的完整性对M-CSF-R的转录激活是必需的。AML1A可以干扰AML1B的转录激活作用。
Objective To investigate the effect of AML1B gene and its isoform AML1A on the transcriptional activity of target gene promoter and to explore the mechanism of hematopoietic stem cell differentiation and leukemia. Methods AML1A and AML1B expression plasmids were constructed and co-transfected with luciferase reporter plasmids containing the target gene macrophage colony-stimulating factor receptor (M-CSF-R) promoter into African green monkey kidney CV-1 cells to measure the fluorescence Enzyme activity, analysis of AML1A and AML1B on target gene transcriptional activity. Results AML1B had obvious transactivation effect on M-CSF-R, which was obviously sequence-specific and dose-dependent. AML1A had no transcriptional activation and antagonized AML1B, which markedly decreased the expression of M-CSF-R . Conclusion The structural integrity of the AML1 transcript is essential for the transcriptional activation of M-CSF-R. AML1A can interfere with the transcriptional activation of AML1B.
