目的:探讨美洲大蠊提取物(CⅡ-3)对两株人肺癌细胞(NCI-H446、NCI-H460)的细胞毒活性及细胞周期的影响。方法:采用改良MTT法检测了CⅡ-3对NCI-H446和NCI-H460人肺癌细胞的细胞毒活性,流式细胞术检测CⅡ-3对2株细胞凋亡、坏死率及细胞周期的的影响。结果:CⅡ-3对NCI-H446和NCI-H460细胞的IC50分别为(48.98±6.63)μg.mL-1、(33.00±10.57)μg.mL-1。对NCI-H446和NCI-H460细胞凋亡、坏死率的影响具有一定的剂量依赖性。对NCI-H446细胞,25,50μg.mL-1CⅡ-3诱导细胞的不同时期发生阻滞,75μg.mL-1CⅡ-3主要诱导细胞发生坏死;对NCI-H460细胞,20,80μg.mL-1CⅡ-3主要诱导细胞S期、G2/M期发生阻滞,40μg.mL-1CⅡ-3主要诱导细胞S期发生阻滞。结论:美洲大蠊提取物CⅡ-3对2株人肺癌细胞有一定的细胞毒活性,且对2株细胞的凋亡、坏死率和细胞周期分布有一定的影响。
Objective: To investigate the effect of C Ⅱ-3 on the cytotoxicity and cell cycle of two human lung cancer cells (NCI-H446 and NCI-H460). Methods: The cytotoxic activity of CⅡ-3 on NCI-H446 and NCI-H460 human lung cancer cells was detected by modified MTT method. The effect of CⅡ-3 on the apoptosis, necrosis rate and cell cycle of the two cell lines was detected by flow cytometry . Results: The IC50 of CⅡ-3 on NCI-H446 and NCI-H460 cells were (48.98 ± 6.63) μg.mL-1 and (33.00 ± 10.57) μg.mL-1, respectively. The apoptosis and necrosis rates of NCI-H446 and NCI-H460 cells were dose-dependent. In NCI-H446 cells, the cells were arrested at different times in 25 and 50μg.mL-1CⅡ-3 cells, and necrosis was induced in 75μg.mL-1CⅡ-3 cells. The NCI-H460 cells were treated with 20, 80μg.mL-1CⅡ -3 mainly induced cell S, G2 / M phase block, 40μg.mL-1CⅡ-3 mainly induced cell S phase arrest. CONCLUSION: Periplactin extract CⅡ-3 has certain cytotoxic activity on two human lung cancer cells and has some effects on the apoptosis, necrosis rate and cell cycle distribution of the two strains.
