目的探讨辽宁省与日本流行北京基因型结核分枝杆菌基因的异同。方法选取辽宁省各市结核病防治所经菌种鉴定得到的144株结核分枝杆菌菌株,以及84株来自日本神奈川县卫生研究所的结核分枝杆菌菌株,采用以聚合酶链式反应(PCR)为基础的分型方法鉴定是否为北京基因型结核分枝杆菌。对引物4扩增辽宁省北京基因型和非北京基因型结核分枝杆菌的条带进行双向测序并比较。结果 144株辽宁省结核分枝杆菌中,北京基因型结核分枝杆菌129株(89.58%),非北京基因型结核分枝杆菌7株(4.86%),北京基因型结核分枝杆菌和非北京基因型结核分枝杆菌混合感染8株(5.56%)。引物4扩增辽宁省7株非北京基因型结核分枝杆菌菌株得到308bp条带,扩增129株辽宁省北京基因型结核分枝杆菌菌株,116株(89.92%)北京基因型结核分枝杆菌出现270 bp左右条带。随机对3株辽宁省北京基因型结核分枝杆菌的270 bp左右条带进行双向测序,序列一致,与非北京基因型结核分枝杆菌的308 bp条带序列不同。84株日本神奈川县的结核分枝杆菌菌株中,北京基因型结核分枝杆菌53株(63.10%),非北京基因型结核分枝杆菌31株(36.90%),引物4扩增日本神奈川非北京基因型结核分枝杆菌菌株得到308 bp条带,扩增北京基因型结核分枝杆菌无条带。结论辽宁省以北京基因型结核分枝杆菌菌株流行为主,存在北京基因型和非北京基因型结核分枝杆菌混合感染。辽宁省流行北京基因型结核分枝杆菌的基因与日本流行的北京基因型结核分枝杆菌存在着不同的特点。
Objective To explore the similarities and differences of Mycobacterium tuberculosis genotypes between Beijing and Japan in Liaoning Province. Methods A total of 144 strains of Mycobacterium tuberculosis strains were identified from strains of tuberculosis prevention and control in Liaoning Province and 84 strains of Mycobacterium tuberculosis strains from Kanagawa Institute of Health in Japan were detected by polymerase chain reaction Basic typing method to identify whether the Beijing genotypes of Mycobacterium tuberculosis. The DNA of primer 4 amplified Beijing genotypes of Beijing and non-Beijing genotypes of M. tuberculosis were bi-directionally sequenced and compared. Results Among 144 strains of Mycobacterium tuberculosis in Liaoning Province, 129 (89.58%) were genotypes of Mycobacterium tuberculosis in Beijing, 7 (4.86%) were non-Beijing genotypes of Mycobacterium tuberculosis, Genotypes of Mycobacterium tuberculosis mixed infection 8 (5.56%). Primer 4 amplified 308 bp bands from 7 non-Beijing genotypes of Mycobacterium tuberculosis strains in Liaoning Province, amplified 129 genotypes of Mycobacterium tuberculosis strains in Beijing, 116 (89.92%) genotypes of Mycobacterium tuberculosis in Beijing, A band of 270 bp appears. The 270 bp bands of three genotypes of Mycobacterium tuberculosis in Beijing were sequenced bidirectionally. The sequence was identical with that of the non - Beijing genotype M. tuberculosis 308 bp. Of 84 strains of Mycobacterium tuberculosis in Kanagawa prefecture in Japan, 53 (63.10%) were genotypes of Mycobacterium tuberculosis in Beijing and 31 (36.90%) were non-genotypes of Mycobacterium tuberculosis. Primer 4 was amplified in non-Beijing The genotype M. tuberculosis strain obtained a 308 bp band to amplify the Beijing genotype M. tuberculosis without band. Conclusions The genotypes of Mycobacterium tuberculosis in Beijing are predominant in Liaoning Province, and the mixed genotypes of Beijing genotypes and non-Beijing genotypes of Mycobacterium tuberculosis exist. The prevalence of Mycobacterium tuberculosis genotypes in Beijing of Liaoning province is different from that of Mycobacterium tuberculosis genotype in Japan.
