目的:观测流体剪切应力作用不同时间对鼠破骨细胞碳酸酐酶II(CAⅡ)mRNA表达的影响。方法:对破骨细胞爬片施加2.9dyne/cm2的流体剪切应力,按作用时间不同分为0min、15min、30min、60min和120min组,采用实时荧光定量PCR检测CAⅡmRNA表达水平,采用SPSS12.0软件包对数据进行单因素方差分析。结果:对照组、15min、30min、60min和120min组CAIImRNA表达量分别为7.88±0.09、11.14±0.12、15.83±0.18、1.94±0.02和1.37±0.01(E+5个拷贝数),任何两组间均见显著性差异(P<0.05)。结论:在试验条件下,随着加载时间的增加,CAⅡmRNA表达水平有增加趋势,至30min组达到峰值,随后表达水平有减小趋势,推测流体剪切应力作用下CAⅡmRNA表达水平存在一定时效关系,但具体作用机制有待进一步研究。
OBJECTIVE: To observe the effect of fluid shear stress on the expression of carbachrophenhydramine II (CAⅡ) mRNA in rat osteoclasts at different times. Methods: Fluid shear stress of 2.9 dyne / cm2 was applied to the osteoclast slide. The rats were divided into 0, 15, 30, 60 and 120 min groups according to the different action time. It is presumed that there is a time-dependent relationship between CAⅡmRNA expression level under the fluid shear stress, However, the specific mechanism needs further study.
