神经元胞体在周围神经损伤修复中起着十分重要的作用.周围神经损伤引起的神经元胞体的退化死亡,无疑是影响神经损伤后功能恢复的一处重要因素.本研究用30只成年Wistar大鼠为实验动物.在手术切断单侧坐骨神经后,于神经近端会接一容积50μl的硅胶囊.分3组分别于囊内充入神经生长因子(20μg)、抗神经生长因子抗体(1:250)和0.9%生理盐水.术后30天取双侧L_4~L_6脊神经节,冰冻连续切片、染色后行神经元计数,并以零维取样随机每组选取100个神经元,测算其形态参数(直径、面积和体积).结果表明,在坐骨神经切断后用生理盐水处理组(空白对照),损伤侧L_4~L_6脊神经节细胞死亡率为29%;在抗神经生长因子抗体组,死亡率为36%;在用神经生长因子处理处,神经元胞体死亡率为2%.形态学测量结果表明,坐骨神经切断术后局部连续应用神经生长因子,神经元各项形态学参数与正常侧近似,无显著差异性;在生理盐水组和抗神经生长因于抗体,神经元均出现明显的萎缩(与正常侧比较,P<0.05).上述结果说明神经生长因子对创伤性感觉神经元的退化死亡有显著的保护作用.文中还就神经生长因子保护创伤性感觉神经元免遭死亡的机制及临床应用的可行性和应用前景等方面提出了看法.
Neuronal soma plays an important role in the repair of peripheral nerve injury.Degraded death of neuronal soma caused by peripheral nerve injury is undoubtedly an important factor affecting the functional recovery after nerve injury.In this study, 30 adult Wistar large Rats were experimental animals.After the operation of unilateral sciatic nerve cut off, a volume of 50μl silicone capsule was taken in the proximal nerve to divide the three groups with nerve growth factor (20μg) and anti-nerve growth factor antibody (1: 250) and 0.9% saline.After 30 days of operation, bilateral L_4 ~ L_6 spinal ganglia were taken and serial sections were frozen and stained for neuronal counts. 100 neurons were randomly selected from each group with zero-dimensional sampling to calculate the morphological parameters (Diameter, area and volume) .Results showed that after the sciatic nerve was cut off, the mortality of L 4 ~ L 6 spinal ganglion cells was 29% in the saline treated group (blank control); in the anti-nerve growth factor antibody group, the mortality was 36%, while in the treatment of nerve growth factor, the death rate of neuronal somatic cell was 2% .Morphological measurement results showed that the local application of nerve growth factor after sciatic nerve transection, the morphological parameters of neurons and positive There was no significant difference between the two groups (P> 0.05) .Normal neurons showed significant atrophy due to antibodies and neurons in normal saline group and anti-NGF (P <0.05) .The above results showed that NGF could inhibit traumatic sensory neurons Of the degenerative death have a significant protective effect.The article also made a point of view on the mechanism of nerve growth factor in protecting traumatic sensory neurons from death and the clinical application of the feasibility and application prospects.
