探讨补肾益气活血方对胎儿宫内生长迟缓(IUGR)胎盘微循环障碍的影响。方法:应用NADPH一黄递酶组织化学方法检测胎盘组织一氧化氮合酶(NOS)活性,采用镇铜镉还原法及硫代巴比妥酸比色法(TBA法)测定了新生儿脐血血浆亚硝酸盐/硝酸盐(NO2-/NO3-)和丙二醛(MDA)浓度。结果:IUGR孕妇胎盘组织NOS活性较正常足月妊娠明显下降,新生儿脐血NO2-/NO3-低于正常新生儿(<0.01),而MDA水平高于正常(P<0.01),中药治疗后与正常对照组差异无显著性(P均>0.05),相关性分析表明NO2-/NO3-与MDA呈明显负相关(r=-0.6124,P<0.05)。结论:脂质过氧化可能是引起IUGR胎盘微循环障碍的中介因子之一,补肾益气活血方促进胎儿宫内生长发育与其抗自由基损伤,增强胎盘组织一氧化氮(NO)合成和分泌有关。
To explore the effect of Bushen Yiqi Huoxue Recipe on placental microcirculatory disturbance of intrauterine growth retardation (IUGR). METHODS: Nitric oxide synthase (NOS) activity in placental tissue was detected by NADPH-diaphorase histochemical method, and umbilical cord blood was measured by town copper cadmium reduction method and thiobarbituric acid colorimetric method (TBA method). Plasma nitrite/nitrate (NO2-/NO3-) and malondialdehyde (MDA) concentrations. Results: The NOS activity of placental tissue in IUGR pregnant women was significantly lower than that in normal term pregnancy. The level of NO2-/NO3- in umbilical cord blood of newborns was lower than that of normal newborns (<0.01), but the MDA level was higher than normal (P<0.01). There was no significant difference between the traditional Chinese medicine treatment group and the normal control group (P>0.05). The correlation analysis showed that there was a significant negative correlation between NO2-/NO3- and MDA (r=-0.6124, P<0.05). . CONCLUSION: Lipid peroxidation may be one of the mediators of IUGR’s placental microcirculatory disturbance. Bushen Yiqi Huoxue Decoction promotes intrauterine growth and development and is associated with anti-free radical injury and enhanced synthesis and secretion of nitric oxide (NO) in placental tissue. .
