目的探讨化疗药物作用下胰腺癌细胞株蛋白质谱的变化,寻找差异表达的蛋白质,为临床化疗提供理论依据。方法利用蛋白质组学技术,观察胰腺癌细胞株SW-1990经吉西他滨和5-氟尿嘧啶(5-FU)处理后,其蛋白质谱的变化,鉴定其差异表达的蛋白质。结果培养细胞受抑制约50%时,吉西他滨为1-100ng/ml。5-FU为250-2500 ng/ml。质谱鉴定了6个差异表达的蛋白质,对照组1个,加药组5个。结论对细胞株的作用,吉西他滨明显优于5-FU。吉西他滨参与了糖代谢和酯类代谢。β-肌动蛋白、MGC:19713、KIAA1058蛋白等某些高表达的差异蛋白质可能成为观察化疗效果的重要指标。
Objective To investigate the changes of protein profiles of pancreatic cancer cell lines under the influence of chemotherapeutic drugs and to search differentially expressed proteins to provide a theoretical basis for clinical chemotherapy. Methods The proteomic changes of pancreatic cancer cell line SW-1990 after treatment with gemcitabine and 5-fluorouracil (5-FU) were observed by proteomic techniques to identify differentially expressed proteins. Results When cultured cells were inhibited by about 50%, gemcitabine was 1-100 ng / ml. 5-FU is 250-2500 ng / ml. Six differentially expressed proteins were identified by mass spectrometry, one in the control group and five in the dosing group. Conclusion The effect of gemcitabine on cell lines is obviously better than that of 5-FU. Gemcitabine is involved in glucose metabolism and ester metabolism. β-actin, MGC: 19713, KIAA1058 proteins and other high expression of the differential proteins may be an important indicator of the effect of chemotherapy.
