应用多层酶联免疫吸着法(ELISA)来测定被谷象幼虫危害的小麦中肌浆球蛋白的含量,并用家蟋蟀Acheta domestica(L.)脚所提取的肌浆球蛋白标定.以单凭仪表检测法进行肌浆球蛋自试验作为在50克小麦样品中昆虫数目和昆虫发育阶段的函数.对每一昆虫而言,从1龄到4龄,肌浆球蛋白含量从0.1增加到9.0μg,蛹期降为1.9μg,羽化前成虫增加到6.6μg,而从X线检测到的总平均值为5.5μg.这种变化与以前所记述的蛀蚀性昆虫肌浆球蛋白含量相似.这一基础标准相当于所研究的在509小麦中有6.8μg肌浆球蛋白含量.在检测小麦蛀蚀情况和面粉厂验收小麦时,这种酶联免疫吸着技术提供了一种灵敏的检测蛀蚀性害虫的方法,大约是在50g小麦中检测6头昆虫,标准误差为土0.8头昆虫.
Multi-layer enzyme-linked immunosorbent assay (ELISA) was used to determine the content of myosin in wheat harmed by grain elephant larvae and was scored with myosin extracted from the leg of Acheta domestica (L.) This change is similar to that previously described for the irrigating insect myosin content. A base standard equates to 6.8 μg of myosin in the wheat studied at 509.
