目的探讨LKB1基因在肝细胞性肝癌(HCC)中的表达及其与肝细胞癌患者预后的关系。方法采用免疫组化技术检测70例HCC患者肝癌及癌旁组织和20例正常肝组织中LKB1蛋白的表达,采用x2检验分析LKB1表达与临床病理因素的关系。对该组70例患者进行随访,并应用COX比例风险回归模型对其预后影响因素进行评估。结果 LKB1蛋白在HCC组织中的阳性表达率明显低于正常肝组织和癌旁组织(P<0.05)。HCC患者中,LKB1蛋白表达与HCC的分化程度,门静脉侵袭,TNM分期有关(P<0.05)。LKB1阳性表达的患者生存时间比阴性表达的显著延长(P<0.05)。门静脉侵袭、胆管癌栓、TNM分期和LKB1表达是影响HCC预后的独立危险因素(P<0.05)。结论 LKB1蛋白表达是影响HCC患者生存时间的独立因素之一,检测LKB1蛋白可能为HCC患者预后判断提供参考。
Objective To investigate the expression of LKB1 in hepatocellular carcinoma (HCC) and its relationship with the prognosis of hepatocellular carcinoma (HCC). Methods Immunohistochemistry was used to detect the expression of LKB1 protein in 70 HCC patients with hepatocellular carcinoma and its adjacent tissues and 20 normal liver tissues. The relationship between LKB1 expression and clinicopathological factors was analyzed by x2 test. The 70 patients in this group were followed up, and the prognostic factors were evaluated by COX proportional hazards regression model. Results The positive expression rate of LKB1 protein in HCC tissues was significantly lower than that in normal liver tissues and adjacent tissues (P <0.05). The expression of LKB1 in HCC patients was related to the differentiation of HCC, portal vein invasion and TNM staging (P <0.05). The survival time of LKB1-positive patients was significantly longer than that of negative ones (P <0.05). Portal vein invasion, cholangiocarcinoma, TNM stage and LKB1 expression were independent risk factors for prognosis of HCC (P <0.05). Conclusion The expression of LKB1 is an independent factor affecting the survival time of HCC patients. Detection of LKB1 protein may provide a reference for the prognosis of HCC patients.
