目的探讨乌头类生物碱(四逆汤煎剂)对体外循环心肌CuZnSOD基因表达的影响。方法28例成年瓣膜置换患者随机分成对照组14例和乌头类生物碱口服(四逆汤煎剂)组14例。术前1周乌头类生物碱组依照中医辩证给予乌头类生物碱每天1剂口服,术后分别取右心耳组织测心肌组织铜锌超氧歧化酶CuZnSODmRNA含量。提取各组心肌组织总RNA通过逆转录聚合酶链反应(RTPCR),以βactin为内参照对CuZnSOD基因的表达水平进行检测。结果乌头类生物碱(四逆汤煎剂)组(1.310±0.135)CuZnSOD基因表达显著强于对照组(1.060±0.183)。结论乌头类生物碱能上调CuZnSOD基因的表达,发挥对体外循环心肌缺血再灌注损伤的保护作用。
Objective To investigate the effect of Aconitum alkaloids (Sini decoction) on the expression of CuZnSOD gene in cardiomyocytes from cardiopulmonary bypass. Methods Twenty-eight adult patients with valve replacement were randomly divided into control group (14 cases) and aconitum alkaloid oral (Sini Decoction) group (14 cases). One week before operation, Aconitum alkaloids group was given a dose of oral administration of Aconitum alkaloids according to Chinese medicine dialectically, and the right atrial appendage tissue was taken to measure CuZnSOD mRNA content of myocardial tissue copper-zinc superoxide dismutase. The expression of CuZnSOD gene was detected by reverse transcription polymerase chain reaction (RTPCR) and β actin as internal reference. Results The CuZnSOD gene expression in the Aconitum alkaloid (Sini Decoction) group was significantly higher than that in the control group (1.360±0.183) (1.310±0.135). Conclusion Aconitum alkaloids can upregulate the expression of CuZnSOD gene and play a protective role against cardiomyocyte ischemia-reperfusion injury.
