驱铅丸对低浓度铅接触大鼠海马组织氧化损伤及超微结构改变的干预作用

目的:从海马区超微结构及抗氧化损伤等方面探讨低浓度铅接触行为记忆改变及中药驱铅丸的干预机制。方法:32只大鼠随机分为4组:空白组饮双蒸水;其他3组饮水中均添加0.02%醋酸铅,其中中药组按驱铅丸3.5g·kg-1·d-1的剂量2ml/d灌服;Ca-EDTA治疗组(依地酸钠钙加普鲁卡因肌肉注射,50mg·kg-1·d-1,连续注射4d为1个疗程,休息4d后进行下一个疗程,连续治疗7个疗程);病理组不予任何治疗。60d后,进行Morris水迷宫试验,检测全血、脑组织铅含量;检测脑组织MDA、SOD和GSH-Px含量;用电镜检测海马组织超微结构。结果:与空白组比较,病理组血铅、脑铅含量显著增高(P<0.01),与病理组比较中药组脑铅、血铅含量显著降低(P<0.05);Morris水迷宫试验结果提示病理组大鼠空间定位航行能力下降(P<0.05),中药组定位能力较病理组明显提高,具有统计学意义(P<0.001);病理组MDA显著高于空白组(P<0.01);与病理组比较,各治疗组MDA含量均显著降低(P<0.05),与病理组比较,中药组SOD、GSH-Px含量显著增高(P<0.05)。结论:驱铅丸可以改善铅接触大鼠的行为记忆障碍,其机制与驱铅丸改善铅导致的氧化损伤有关。 OBJECTIVE: To investigate the changes in behavior and memory of low-level lead exposure and the intervention mechanism of traditional Chinese medicine and lead pill from the aspects of ultrastructure and antioxidative damage in hippocampus. METHODS: Thirty-two rats were randomly divided into four groups: the control group was treated with double distilled water; the other three groups of drinking water were supplemented with 0.02% lead acetate, and the Chinese medicine group was given a dosage of 2.5g·kg-1·d-1 of 2ml. /d Dose; Ca-EDTA treatment group (sodium edetate calcium plus procaine intramuscular injection, 50mg · kg-1 · d-1, continuous injection 4d for a course of treatment, rest after 4d for the next course of treatment, Seven consecutive courses of treatment); pathological group without any treatment. After 60 days, the Morris water maze test was performed to detect the levels of lead in the whole blood and brain tissues; the contents of MDA, SOD, and GSH-Px in brain tissue were detected; the ultrastructure of hippocampal tissue was detected by electron microscope. Results: Compared with the blank group, the blood lead and brain lead levels in the pathological group were significantly higher (P<0.01). Compared with the pathological group, the brain lead and blood lead levels in the traditional Chinese medicine group were significantly lower (P<0.05); Morris water maze test results suggested pathology The spatial navigation ability of the rats decreased (P<0.05). The positioning ability of the Chinese medicine group was significantly higher than that of the pathological group (P<0.001). MDA in the pathological group was significantly higher than that in the blank group (P<0.01); Compared with the pathological group, the contents of SOD and GSH-Px in the Chinese medicine group were significantly higher (P<0.05). Conclusion: Quxie Pill can improve the behavior and memory impairment in rats exposed to lead. The mechanism is related to the improvement of lead-induced oxidative damage.
