目的评估基于VP4区基因序列的肠道病毒分型方法准确性,建立快速简便的肠道病毒分型方法。方法应用逆转录-聚合酶链反应(RT-PCR)对126株已用VP1区基因序列完成分型鉴定的肠道病毒进行VP4区基因扩增,对扩增产物测序分析,获得VP4区分型结果。结果 126株肠道病毒经VP4区基因序列分型共分出30个型别,包括肠道病毒A组37株8个型别,肠道病毒B组85株20个型别,肠道病毒C组2株2个型别。与VP1区分型结果比较,A、C组肠道病毒所测型别符合率100%;B组肠道病毒所测型别仅有5个符合,符合率21%。结论基于VP4区基因序列的肠道病毒分型方法对A组肠道病毒能准确分型,对B组肠道病毒分型准确率欠佳,C组肠道病毒株较少,分型准确率有待进一步验证。
Objective To evaluate the accuracy of the typing method of enterovirus based on the VP4 region gene sequence and to establish a rapid and simple typing method of enterovirus. Methods The VP4 region of 126 strains of Enteroviruses which had been identified by VP1 gene sequence were amplified by reverse transcription polymerase chain reaction (RT-PCR). The amplified products were sequenced and the VP4 typing results were obtained . Results The results showed that there were 30 genotypes in 126 strains of enterovirus by VP4 gene sequence typing, including 37 strains of enterovirus A, 37 strains of 8 strains, 85 strains of enterovirus B, 20 strains of enterovirus C Group 2 strains 2 types. Compared with the results of VP1 differentiating type, the coincidence rate of type A and group C enterovirus was 100%; only 5 of the group B enterovirus conformed, the coincidence rate was 21%. Conclusions The classification of enterovirus in group A based on the sequence of VP4 gene sequence can accurately classify the group B. The typing accuracy of enterovirus in group B is not good. The number of enteric virus in group C is less, the accuracy of typing To be further verified.
