目的采用超高效液相色谱仪与四极杆飞行时间串联质谱仪联用技术(UHPLC-Q-TOF/MS)对藜芦中化学成分进行分析鉴别。方法应用Agilent Eclipse Plus C18色谱柱(2.1 mm×100 mm,1.8μm),以0.1%甲酸水溶液(A)-乙腈(B)为流动相梯度洗脱,电喷雾离子源,正离子模式下采集数据。结果共鉴别出黎芦药材中32个化学成分,通过比较碎片离子对其中的3对同分异构体进行了结构解析。结论采用UHPLC-Q-TOF/MS方法能快速地在一张图谱上表征出藜芦中的主要化学成分,为藜芦的药效物质基础及化学成分体内代谢与作用机制的深入研究奠定了基础。
OBJECTIVE To analyze and identify the chemical constituents of veratrum by ultra-high performance liquid chromatography coupled with quadrupole time of flight tandem mass spectrometry (UHPLC-Q-TOF / MS). Methods Agilent Eclipse Plus C18 column (2.1 mm × 100 mm, 1.8 μm) was used for the mobile phase gradient elution with 0.1% formic acid (A) -acetonitrile (B). Electrospray ionization (ESI) . Results A total of 32 chemical constituents of Radix Rehmanniae were identified, and three isomers of them were structurally analyzed by comparison of fragment ions. Conclusion UHPLC-Q-TOF / MS method can quickly characterize the main chemical constituents of Veratrum, setting the foundation for the pharmacodynamic basis of Veratrum and the in-depth study of the metabolism and mechanism of chemical constituents in vivo .
