目的研究嗅鞘细胞(OECs)的培养及其对自身免疫性脑脊髓炎(EAE)的治疗作用。方法取36只EAE模型大鼠,将其随机分为移植组18只,假手术组18只。用新生Wistar大鼠嗅球培养出OECs,移植组将OECs悬液注入EAE大鼠的侧脑室;假手术组按以上方法注入等量生理盐水。观察2组大鼠运动功能评分、组织病理学变化以及血管内皮生长因子(VEGF)表达情况。结果(1)OECs移植组运动功能评分从第1天的(6.8±0.7)分降至第20天的(1.1±0.3)分,显著优于假手术组,差异有统计学意义(P<0.05);(2)组织病理切片可见OECs移植组髓鞘修复明显优于假手术组;(3)OECs移植组VEGF的表达从第1天的(22.1±2.8)个/mm2增至第10天的(300.6±5.1)个/mm2,后逐渐降至第21天的(99.0±3.5)个/mm2,在各时间点的表达均明显高于假手术组,差异有统计学意义(P<0.01)。结论OECs移植可以促进EAE的髓鞘修复,改善运动功能,同时可使VEGF大量表达,诱导病灶周围新生血管的生成。
Objective To study the culture of olfactory ensheathing cells (OECs) and its therapeutic effect on autoimmune encephalomyelitis (EAE). Methods Thirty-six EAE rats were randomly divided into 18 rats in transplantation group and 18 rats in sham operation group. The OECs were cultured in the olfactory bulb of newborn Wistar rats. The rats in the transplantation group were injected with OECs suspension into the lateral ventricle of EAE rats. The rats in the sham operation group were injected with the same volume of normal saline. Motor function score, histopathological changes and expression of vascular endothelial growth factor (VEGF) in the two groups were observed. Results (1) The score of motor function in OECs transplantation group decreased from (6.8 ± 0.7) on day 1 to (1.1 ± 0.3) on day 20, which was significantly better than that on sham operation group (P <0.05 ). (2) The histological sections showed that myelin sheath repair in OECs transplantation group was better than sham operation group. (3) The expression of VEGF in OECs transplantation group increased from (22.1 ± 2.8) / mm2 on day 1 to (300.6 ± 5.1) / mm2, and then gradually decreased to (99.0 ± 3.5) / mm2 on the 21st day, and the expression at each time point was significantly higher than that in sham operation group (P <0.01) . Conclusion OEC transplantation can promote myelin sheath repair of EAE and improve motor function, meanwhile, it can express large amount of VEGF and induce neovascularization around the lesion.
