目的探讨魔芋多糖对高脂饮食小鼠血清瘦素和小肠粘膜Na+-K+-ATP酶水平的影响。方法将实验动物分为正常对照组、高脂对照组和高、中、低剂量魔芋多糖与高脂联合处理组共5个组,连续喂饲20天,用ELISA法测定血清瘦素水平,分光光度法测定小肠粘膜Na+-K+-ATP酶活性以及体重、体脂、血糖等指标。结果第10天高脂对照组的体重和餐后血糖为(31.3±2.11)g和(7.5±1.15)mmol/L,魔芋多糖高剂量组则为(28.0±2.06)g和(4.8±0.73)mmol/L。第20天高脂对照组的血清瘦素浓度和小肠粘膜Na+-K+与高脂联合处理-ATP酶活性为(1078.5±61.69)pg/ml和(16.2±1.48)μmolPi/(mg pro·h),魔芋多糖高剂量与高脂联合处理组则为(820.5±58.52)pg/ml和(11.2±1.10)μmolPi/(mgpro·h)。两组间各项指标差别均有显著性意义(P<0.05)。结论魔芋多糖能够降低高脂饮食小鼠餐后血糖、血清瘦素水平和小肠粘膜Na+-K+-ATP酶活性,抑制体重和体脂的增加。
Objective To investigate the effects of konjac polysaccharide on the levels of serum leptin and Na+-K+-ATPase in murine small intestine mucosa with high fat diet. Methods The experimental animals were divided into normal control group, high-fat control group and high-, middle-, and low-dose konjac polysaccharides and high-fat combination treatment groups. Five groups were continuously fed for 20 days. Serum leptin levels were measured by ELISA method. Spectrophotometric determination of Na+-K+-ATPase activity in the small intestine mucosa and indicators of body weight, body fat, and blood glucose. Results On day 10, the body weight and postprandial blood glucose of the high-fat control group were (31.3±2.11) g and (7.5±1.15) mmol/L, and those of the high-dose konjac polysaccharide group were (28.0±2.06) g and (4.8±0.73). Mmol/L. On the 20th day, the serum leptin concentration in the high-fat control group and the combination of Na+-K+ and high-lipid in the small intestine mucosa-ATPase activity were (1078.5±61.69) pg/ml and (16.2±1.48) μmol Pi/(mg pro·h). The high-dose and high-fat combination treatment of konjac polysaccharide was (820.5±58.52) pg/ml and (11.2±1.10) μmolPi/(mgpro·h). Significant differences were found between the two groups (P<0.05). Conclusion Konjac polysaccharide can reduce postprandial blood glucose, serum leptin levels, and Na+-K+-ATPase activity of small intestine mucosa in mice with high-fat diet, and inhibit the increase of body weight and body fat.
