结合区段灌注技术,以万古霉素为手性选择剂构建毛细管电泳手性分离体系,研究了氨基酸对映异构体的分离。选择的最佳分离条件为:①含3mmol.L-1万古霉素和2mmol.L-1HDB的pH 7为200mmol.L-1硼酸缓冲溶液的分离区段;②灌注长度为压力3.45kPa,时间2.5min。以含2mmol.L-1 HDB的pH 7为200mmol.L-1硼酸缓冲溶液为运行缓冲溶液对19种氨基酸衍生物进行手性分离。试验结果显示其中15对氨基酸对映异构体得到了良好分离。
Combined with zone perfusion technique, chiral separation system was established by capillary electrophoresis with vancomycin as a chiral selective agent to study the separation of amino acid enantiomers. The optimal separation conditions were as follows: ① separation section containing 200 mmol.L-1 borate buffer solution containing 3 mmol.L-1 vancomycin and 2 mmol.L-1HDB; ② the perfusion length was 3.45 kPa, 2.5min. Nineteen kinds of amino acid derivatives were chirally separated by using 200 mmol. L-1 borate buffer solution containing 2 mmol.L-1 HDB as the running buffer solution. The test results showed that 15 pairs of amino acid enantiomers were well separated.
