目的采用超高效液相色谱法测定桑叶提取物中槲皮素和山柰酚的含量。方法采用Agilent Ecilipse C18色谱柱(50mm×2.1 mm,1.8-Micron),以乙腈-水(含0.1%甲酸)(35∶65)为流动相,流速为0.4 m L·min-1,柱温为25℃,检测波长370 nm,以外标法定量,进样量2.0μL。结果槲皮素和山柰酚0.01~1.50 mg·m L-1与峰面积的线性关系良好,平均回收率分别为100.11%、101.92%,RSD分别为0.10%、0.16%。结论该测定方法快速,准确,可用于桑叶提取物中槲皮素和山柰酚的含量测定。
Objective To determine the contents of quercetin and kaempferol in mulberry leaves extract by ultra performance liquid chromatography. Methods The mobile phase was acetonitrile-water (containing 0.1% formic acid) (35:65) with a flow rate of 0.4 m L · min-1 on an Agilent Ecilipse C18 column (50 mm × 2.1 mm, 1.8-Micron) 25 ℃, detection wavelength 370 nm, external standard method, injection volume 2.0μL. Results The linear relationship between quercetin and kaempferol at 0.01-1.50 mg · m L-1 and peak area was good. The average recoveries were 100.11% and 101.92%, respectively, with RSDs of 0.10% and 0.16%, respectively. Conclusion The method is rapid and accurate and can be used for the determination of quercetin and kaempferol in mulberry leaves extract.
