培养正常人造血组织的红系集落惯用甲基纤维素(MC)或血浆凝块法。用MC的主要弊端是不能制备用于细胞学或细胞化学分析的永久性标本。本研究的基本目的是:确定琼脂是否象MC一样有效地支持正常人红系细胞的体外生长,其次是通过缩小培养体系来节省红细胞生成素(Epo)。 MC培养依据Guilbert和Iscove的方法。其培养体系为:含终浓度30%的胎牛血清(FBS)的Iscove改良的Dulbecco培养基(IMDM)。Epo的终浓度为2u/ml,细胞浓度为2×10~5/ml。全部实验用同一批
Red blood cells cultured in normal human hematopoietic tissue are often treated with methylcellulose (MC) or plasma clotting. The major drawback of using MC is the inability to prepare permanent specimens for cytological or cytochemical analysis. The basic purpose of this study was to determine whether agar effectively supports the growth of normal human erythroid cells in vitro as well as MC, and secondly to conserve erythropoietin (Epo) by reducing the culture system. MC training is based on the method of Guilbert and Iscove. The culture system was: Iscove’s Modified Dulbecco’s Medium (IMDM) with a final concentration of 30% fetal bovine serum (FBS).
