目的探讨重组人粒细胞集落刺激因子(rhG-CSF)对缺血大鼠神经生长因子蛋白表达的影响,并观察糖尿病对其影响。方法应用线栓法制作糖尿病及非糖尿病大鼠大脑中动脉栓塞模型后,将大鼠随机分为两组,治疗组连续sc rhG-CSF(50μg·kg-1·d-1),对照组注射等量生理盐水,进行神经功能缺损评分;计数神经细胞凋亡数量,检测NGF蛋白的表达。结果治疗组大鼠的神经功能缺损评分及缺血周边区域的TUNEL阳性细胞数均低于对照组的(P<0.01),而神经生长因子蛋白免疫阳性细胞多于对照组的(P<0.01);非糖尿病组较糖尿病组更明显。结论 rhG-CSF可增加缺血性脑卒中后周边区域的神经生长因子蛋白的表达,同时抑制神经细胞凋亡,最终改善神经功能预后;糖尿病可加重脑梗死。
Objective To investigate the effect of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on the expression of nerve growth factor (NGF) protein in rats with ischemia and to observe the effect of diabetes on it. Methods The middle cerebral artery occlusion model of diabetic and non-diabetic rats was made by thread method. The rats were randomly divided into two groups. The treatment group received continuous rhG-CSF (50μg · kg-1 · d-1) The same amount of saline, neurological deficit score; count the number of nerve cell apoptosis, detection of NGF protein expression. Results The neurological deficit score and the number of TUNEL-positive cells in the peripheral area of the ischemic area in the treatment group were lower than those in the control group (P <0.01), while the number of NGF positive cells was higher in the treatment group than in the control group (P <0.01) ; Non-diabetic group more obvious than the diabetic group. Conclusion rhG-CSF can increase the expression of nerve growth factor protein in peripheral region after ischemic stroke, inhibit the apoptosis of nerve cells and finally improve the prognosis of neurological function. Diabetes mellitus can aggravate cerebral infarction.
