目的建立以反相高效液相色谱-蒸发光散射检测法(RP-HPLC-ELSD)测定不同产地蕤仁中亚油酸含量的测定方法,为有效控制蕤仁饮片的质量提供科学依据。方法采用回流法提取亚油酸;色谱条件为:AichromaBond-AQ C18色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.1%甲酸水(90∶10);流速:1.0 mL·min-1;柱温:25℃;ELSD检测器。结果亚油酸对照品在0.998 9~49.945μg内线性关系良好(r=0.998 4),平均回收率为100.1%,RSD为1.2%。结论本方法准确,重复性好,简便易行,适用于蕤仁中亚油酸的含量测定。
OBJECTIVE To establish a method for the determination of linoleic acid in Rumex glabra by RP-HPLC-ELSD and to provide a scientific basis for the effective control of the quality of Rumex japonicus. Methods The refluxing method was used to extract linoleic acid. The chromatographic conditions were as follows: AichromaBond-AQ C18 column (4.6 mm × 250 mm, 5 μm); mobile phase: acetonitrile-0.1% formic acid water -1; column temperature: 25 ° C .; ELSD detector. Results The linoleic acid reference substance had good linearity (r = 0.998 4) in 0.998 9 ~ 49.945 μg with an average recovery of 100.1% and a RSD of 1.2%. Conclusion This method is accurate, reproducible, simple and easy to use, suitable for the determination of linoleic acid linoleate content.
