目的观察毛冬青对心衰大鼠心功能及微小RNA133a(miR133a)表达的影响。方法腹主动脉缩窄术复制大鼠心衰模型,随机分为毛冬青高、低剂量组,卡托普利组,心衰模型组和假手术组,干预2周后抽血查B型尿钠肽(BNP)和行心脏彩超检查心功能,并对大鼠心脏组织miR133a进行荧光PCR检测。结果心衰模型组的心功能指标明显低于假手术组,3个药物干预组心功能指标均有较好的提高作用,组间作用无显著性差异;与假手术组比较,心衰模型组miR133a的表达下调,各药物干预组与心衰模型组比较,miR133a的表达均显著上升,其中以毛冬青低剂量组最为显著。结论腹主动脉缩窄可成功复制大鼠慢性心力衰竭模型,心力衰竭大鼠模型较假手术组大鼠miR133a的表达下调。不同剂量的毛冬青均能改善慢性心力衰竭大鼠模型的心功能,并且能上调miR133a的表达,但上调幅度与心功能改善情况未呈明显量效关系。
Objective To observe the effect of Mao dongqing on the cardiac function and the expression of miR133a in heart failure rats. Methods A rat model of heart failure was established by abdominal aorta constriction. The rats were randomly divided into two groups: high-dose and low-dose group, captopril group, heart failure model group and sham-operation group. BNP and cardiac ultrasonography were used to examine cardiac function. Fluorescent PCR was performed on miR133a in rat cardiac tissue. Results Compared with the sham-operated group, the cardiac function index of the model group was significantly lower than that of the sham-operated group. The cardiac function indexes of the three drug-intervention groups all had a good effect. There was no significant difference between the two groups. Compared with the sham operation group, The expression of miR133a was down-regulated. Compared with the model group, the expression of miR133a in each drug intervention group was significantly increased, especially in Mao Dongqing low-dose group. Conclusions Abdominal aortic constriction can successfully replicate the model of chronic heart failure in rats. The expression of miR133a in heart failure rat model is lower than that in sham operation group. Different doses of Mao Dongqing can improve the heart function of chronic heart failure rat model, and can up-regulate miR133a expression, but the rate of increase and cardiac function did not show a significant dose-effect relationship.
