目的 :了解伯氨喹 (伯喹 )对大鼠高铁血红蛋白 ( MHb)生成和网织红细胞 ( ret)增生的影响 ,并与苯胺所致的大鼠急性溶血反应进行比较。方法 :SD大鼠经腹腔 ( ip) 1次注射 3%苯胺花生油溶液 1ml/ kg,于给药前和给药后 2 h、2 4 h、4 8h和 72 h分别取血测定 MHb和 ret;另 1组大鼠灌胃 ( ig)伯喹 4 0 mg/ kg· d× 7d,于给药前及首剂给药的次日起 ,逐日取血检测 MHb和 ret直至停药后第 5d,以后隔日血检至停药后 15d。以上 2个给药组均设相应的溶剂对照。另对 37只大鼠进行了 G6PD活力测定。结果 :苯胺组给药后 2 h,MHb即显著升高 ,2 4 h恢复正常 ;ret自给药后 2 4 h开始上升 ,至 72 h仍持续升高 ,表现为急性血管内溶血反应。伯喹组给药期间及停药后MHb未见明显变化 ;ret于给药的 d5起持续低于对照组 ,至停药后的 d3— d5反应性增高 ,以后逐渐恢复正常。大鼠的 G6PD活力为 4 2 .3± 11.1%。结论 :伯喹对大鼠血液系统的影响表现为抑制网织红细胞的增生 ,但不引起类似苯胺所致的急性溶血反应。
Objective: To investigate the effect of primaquine on the formation of methemoglobin (MHb) and the proliferation of reticulocytes in retina in rats, and to compare with the acute hemolytic reaction induced by aniline in rats. Methods: SD rats were intraperitoneally injected with 1 ml / kg of 3% aniline peanut oil solution 1 hour before administration and at 2, 24, 48 and 72 h after administration. Another group of rats were intragastrically administered ig with primary quinone 40 mg / kg · d × 7d. Blood samples were taken daily for detection of MHb and ret day before drug administration and on the 5th day after drug withdrawal, After the next day blood test to 15d after withdrawal. The above two groups were given the corresponding solvent control. Another 37 rats were G6PD activity determination. Results: At 2 h after administration, the MHb level increased significantly and returned to normal after 24 h. Ret began to increase at 24 h and continued to increase at 72 h. The acute intravascular hemolysis was observed. No significant change of MHb was found during the administration of berberine and d3-d5 after withdrawal from d5 on d5, which was gradually lower than that of the control group after retreatment, and returned to normal afterwards. The G6PD activity of rats was 42.3 ± 11.1%. CONCLUSION: The effect of primaquine on the blood system in rats is inhibited by reticulocyte proliferation, but does not cause aniline-induced acute hemolytic reaction.
