目的分析乙型肝炎病毒基因组中增强子Ⅰ区的特征,进一步探讨乙肝慢性化的机制。方法收集16份慢性乙型肝炎患者的血清标本HBV-DNA,扩增HBV EnhⅠ基因,PCR产物测序后对HBV进行分型,并分析HBVEnhⅠ突变情况。结果 10例(62.5%)标本HBV EnhⅠ基因PCR扩增阳性,测序标本经在线Genotyping比对后,均属于B型,与参考序列AF100309同源性最高,10份标本的HBV EnhⅠ区共发现12个突变位点。结论 HBVEnhⅠ基因存在多位点突变,此可能为促进病毒复制导致乙肝慢性化的机制。
Objective To analyze the characteristics of enhancer Ⅰ region in hepatitis B virus genome and further explore the mechanism of hepatitis B chronicity. Methods Serum samples of HBV-DNA from 16 patients with chronic hepatitis B were collected and HBV Enh Ⅰ gene was amplified. HBV was sequenced after PCR products were sequenced, and HBVEnh Ⅰ mutation was analyzed. Results The HBV Enh Ⅰ gene was positive in 10 cases (62.5%). The sequence of HBV Enh Ⅰ gene was type B after in-line Genotyping, which had the highest homology with reference sequence AF100309. Twelve HBV Enh Ⅰ regions were found in 10 specimens Mutation site. Conclusion There are multiple site mutations in HBVEnh Ⅰ gene, which may be the mechanism of promoting chronic hepatitis B virus replication.
